Organotypic Cortex Cultures

Organotypic cortex cultures are ex vivo brain-slice preparations maintained in vitro while preserving key features of cortical architecture, including local neuronal and glial interactions. Typically, thin cortical sections are placed under controlled nutrient, temperature, and gas conditions, allowing cells and neural circuits to remain viable and develop over time with limited disruption of tissue organization. These cultures provide an accessible model for studying synaptic connectivity, neuronal development, neurodegeneration, injury responses, and circuit physiology. They also support experimental manipulation and imaging, helping researchers investigate cortical mechanisms while reducing the complexity and variability of whole-animal experiments.

Organotypic Hippocampal Slice Cultures

We describe a method to prepare organotypic hippocampal slices that can be easily adapted to other brain regions. Brain slices are laid on porous membranes and culture media is allowed to form an interface. This method preserves the gross architecture of the hippocampus for up to 2 weeks in culture.

A Model of Epileptogenesis in Rhinal Cortex-Hippocampus Organotypic Slice Cultures

Here, we describe the preparation of rhinal cortex-hippocampus organotypic slices. Under a gradual and controlled deprivation of serum, these slices depict evolving epileptic-like events and can be considered an ex vivo model of epileptogenesis. This system represents an excellent tool for monitoring the dynamics of spontaneous activity, as well as for assessing the progression of neuroinflammatory features throughout the course of epileptogenesis.
