Postnatal Brain Development

Postnatal brain development is the maturation of the brain after birth, a dynamic process that shapes neural circuits, behavior, and cognitive function. It involves coordinated synaptic formation and pruning, myelination, changes in neurotransmitter systems, and activity-dependent plasticity, through which experience and biological signals refine connections among neurons. In neuroscience, studying these changes helps explain sensitive periods in learning, language, and sensory development, while differences in developmental timing can inform research on neurodevelopmental disorders. Longitudinal imaging, electrophysiology, and molecular analyses provide complementary ways to connect cellular mechanisms with emerging brain function.

Mosaic Analysis of Gene Function in Postnatal Mouse Brain Development by Using Virus-based Cre Recombination

An in vivo method to test gene function in postnatal brain is described. Recombinant AAVs expressing Cre and/or a fluorescent protein are injected into neonatal mouse brain. Mosaic gene inactivation and sparse neuronal labeling are achieved, allowing rapid analysis of gene function in processes critical to neural circuit development.

Neuroblast migration is a fundamental event in postnatal neurogenesis. We describe a protocol for efficient labeling of neuroblasts by in vivo postnatal electroporation and subsequent visualization of their migration using time-lapse imaging of acute brain slices. We include a description for the quantitative analysis of neuroblast dynamics by video tracking.
