Stereotaxic Plane Maintenance

Stereotaxic plane maintenance is the process of keeping an animal’s skull and brain in a defined three-dimensional orientation during neurosurgical procedures, which supports accurate targeting and reproducible coordinates. It works by stabilizing the head with ear bars and a bite bar, then adjusting the apparatus until anatomical landmarks such as bregma and lambda are aligned in the same reference plane; this minimizes angular error as instruments advance. Maintaining this plane is essential for intracranial injections, electrode implantation, lesioning, and other procedures that depend on stereotaxic atlases. Consistent alignment improves placement accuracy, experimental comparability, and interpretation of neural circuitry and behavior.

Source: Kay Stewart, RVT, RLATG, CMAR; Valerie A. Schroeder, RVT, RLATG. University of Notre Dame, IN The Guide for the Care and Use of Laboratory Animals ("The Guide") states that pain assessment and alleviation are integral components of the veterinary care of laboratory animals. The definition of anesthesia is the loss of feeling or sensation. It is a dynamic event involving changes in anesthetic depth with respect to an animal's metabolism, surgical stimulation, or variations in the...

The monitoring of extracellular neurotransmitter levels in distinct brain regions of freely moving animals offers insights on the link between neurotransmitter release and behavior. In vivo microdialysis coupled with electrochemical detection provides excellent anatomical and chemical resolution; and information on how basal neurotransmission is altered by pharmacological or physiological manipulations.

Stereotaxic (or stereotactic) surgery is a method used to manipulate the brain of living animals. This technique allows researchers to accurately target deep structures within the brain through the use of a stereotaxic atlas, which provides the 3D coordinates of each area with respect to anatomical landmarks on the skull. After the skull is exposed, anesthetized animals are mounted on a specialized instrument known as a stereotaxic frame, which enables the precise placement of experimental...

Drosophila Maintenance

Drosophila melanogaster, commonly known as fruit flies, are a frequently used model organism for life science research. Although starting a collection of these critters may seem as easy as leaving a banana on your kitchen counter for too long, a productive fly colony in the lab requires careful husbandry and maintenance. This video demonstrates the necessary steps for maintaining a healthy fly stock. The overview begins with the preparation and storage of the yeast and sugar-containing media on...
