Ejection Fraction

Ejection fraction is the percentage of blood pumped out of a ventricle with each heartbeat, providing a standard measure of cardiac systolic function. It is calculated by subtracting end-systolic volume from end-diastolic volume, dividing the difference by end-diastolic volume, and expressing the result as a percentage, commonly using measurements from echocardiography. In pharmacology, ejection fraction helps characterize heart failure, distinguish reduced from preserved ventricular function, and assess responses to therapies that alter contractility, preload, or afterload. Tracking changes over time supports drug evaluation, treatment selection, and monitoring of cardiac effects during clinical research.

This work introduces two computational models of heart failure with preserved ejection fraction based on a lumped-parameter approach and finite element analysis. These models are used to evaluate the changes in the hemodynamics of the left ventricle and related vasculature induced by pressure overload and diminished ventricular compliance.

Here, we describe an optimized, Langendorff-based procedure for the isolation of single-cell atrial cardiomyocytes from a rat model of metabolic syndrome-related heart failure with preserved ejection fraction. A manual regulation of intraluminal pressure of cardiac cavities is implemented to yield functionally intact myocytes suitable for excitation-contraction-coupling studies.
