Les inhibiteurs de transcription sont capables de réprimer la transcription d'un gène. Les inhibiteurs peuvent également se lier à leurs propres séquences inhibitrices cis-régulatrices, empêchant ainsi l'initiation de la transcription même en présence d'un activateur sur la région de régulation. L'un des mécanismes les plus couramment utilisés par les inhibiteurs de transcription est d'utiliser son domaine répresseur pour recruter des protéines appelées co-répresseurs.Les co-répresseurs ont souvent des activités enzymatiques qui peuvent inhiber la transcription. De nombreux co-répresseurs fonctionnent comme des histones désacétylases, des histones méthyltransférases et d'autres agents de remodélisation de la chromatine. Eukaryotic Transcription Inhibitors

Certains processus biochimiques, comme le développement embryonnaire et la régulation de la croissance cellulaire, dépendent de la répression de gènes spécifiques. Les protéines de liaison à l'ADN, connues sous le nom d'inhibiteurs de transcription eucaryotes, régulent la répression de l'expression des gènes chez les eucaryotes. La présence de ces inhibiteurs à l’endroit et au moment requis dans la cellule est déclenchée par la présence d’hormones et de signaux supplémentaires provenant d’autres cellules.

Les inhibiteurs de transcription eucaryotes contiennent généralement deux domaines distincts, un domaine de liaison à l'ADN et un domaine répresseur. Les domaines répresseurs des inhibiteurs de transcription peuvent se lier à d'autres protéines, telles que les régulateurs transcriptionnels basaux, les activateurs, les co-activateurs et les co-répresseurs. Les domaines de répression peuvent être vaguement classés sur la base de leurs acides aminés constitutifs, tels que les domaines riches en alanine, les domaines riches en glutamine et les domaines riches en proline. Ils peuvent également être classés en domaines acides ou basiques, et en domaines hydrophiles ou hydrophobes. Ces domaines sont souvent interchangeables et différents domaines répresseurs peuvent s'associer au même domaine de liaison à l'ADN.

Les inhibiteurs de transcription eucaryotes peuvent remplir leur fonction par le biais de trois mécanismes principaux : l'inhibition de la liaison à l'ADN, le blocage de l'activation et l'inactivation des gènes.

L'inhibition de la liaison à l'ADN empêche l'activateur de se lier au site promoteur ou amplificateur, ou peut empêcher le complexe d'initiation de la transcription de se lier au site d'initiation de la transcription ou à la boîte TATA. Cela pourrait se produire soit par la liaison de l'inhibiteur au site régulateur ou d'initiation, soit par l'association du domaine inhibiteur avec des activateurs, des co-activateurs ou des protéines impliquées dans la machinerie de transcription. Par exemple, chez l'homme, l'inhibiteur de transcription NF-E se lie à la boîte CCAAT du gène de la γ-globine fœtale et inhibe la liaison de l'activateur CP1.

Le blocage de l'activation empêche le signal d'un activateur de transcription lié à un promoteur ou à un amplificateur d'atteindre le site d'initiation de la transcription. Cela peut se produire lorsqu'un inhibiteur se lie à un site adjacent au promoteur ou à l'amplificateur, comme dans la régulation de c-myc où l'inhibiteur myc-PRF se lie à côté du promoteur. Cela peut également se produire par la liaison de l’inhibiteur à un activateur lié à l’ADN. Ce type de répression se produit lorsque l'inhibiteur GAL80 de la levure se lie à l'activateur GAL4 lorsqu'il est lié au promoteur.

Le silençage génique se produit lorsque l'inhibiteur verrouille ou désorganise le complexe d'initiation de la transcription en se liant simultanément à l'une des protéines qui constituent la machinerie et à un site de l'ADN proche du site d'initiation de la transcription, ancrant ainsi la machinerie de transcription. Par exemple, chez les levures, l'inhibiteur α-2 inactive les gènes spécifiques du type sexuel a en se liant sous forme de dimère à l'activateur GRM et aux séquences d'ADN bordant le site de liaison du GRM.

Eukaryotic gene repressors help turn specific genes off when the cell needs to control growth, development, or other biochemical processes. These DNA-binding proteins regulate gene expression in eukaryotes. Their presence at the right time and place can be triggered by hormones and signals from other cells.

Most transcription inhibitors have two main parts. One part binds DNA, and the other is a repressor domain, which helps shut down transcription. Repressor domains can connect with basal transcription regulators, activators, co-activators, and co-repressors.

These repression domains are often grouped by their amino acids, such as alanine-rich, glutamine-rich, and proline-rich domains. They can also be described as acidic or basic, and as hydrophilic or hydrophobic. Different repressor domains can often be swapped onto the same DNA-binding domain.

Eukaryotic transcription inhibitors act through three main mechanisms: inhibiting DNA binding, blocking activation, and gene silencing. Inhibiting DNA binding stops an activator from attaching to a promoter or enhancer, or it prevents the transcription initiation complex from binding the transcription start site or the TATA box. This can happen when the inhibitor binds the regulatory site or start site, or when its inhibitory domain interacts with activators, co-activators, or other transcription machinery proteins.

One example is NF-E in humans. It binds the CCAAT box of the fetal gamma-globin gene and blocks the activator CP1 from binding. Blocking activation works differently. The inhibitor can bind next to a promoter or enhancer, as in c-myc regulation, where myc-PRF binds beside the promoter. It can also bind to an activator already attached to DNA. In yeast, GAL80 binds GAL4 when GAL4 is bound to the promoter.

Gene silencing happens when the inhibitor locks or disrupts the transcription initiation complex. It does this by binding both a protein in the machinery and a DNA site near the transcription start site, which anchors the complex. In yeast, the alpha-2 inhibitor works this way to turn off mating type a-specific genes. It binds as a dimer to the activator GRM and to DNA sequences beside the GRM binding site.
