DNA Isolation

L’ADN doit être isolé des cellules et coupé à des positions précises pour de nombreuses applications, telles que la technologie de l’ADN recombinant.
Bien que différents types de méthodes d’extraction d’ADN soient utilisés pour différents types de cellules, il existe trois étapes standard : la lyse cellulaire, l’élimination des protéines et la récupération de l’ADN.
À l’intérieur des cellules eucaryotes, l’ADN est emballé dans le noyau. Ainsi, la membrane cellulaire et la membrane nucléaire doivent être rompues pour isoler l’ADN.
Cette étape peut être effectuée mécaniquement en décomposant les cellules par broyage ou sonication, ou chimiquement en utilisant des détergents et des enzymes pour dissoudre des parties de la membrane cellulaire.
Une fois le contenu de la cellule libéré, les débris sont séparés des composants solubles par centrifugation. Le surnageant récupéré contient des acides nucléiques et des protéines hydrosolubles.
Pour éliminer les protéines, des enzymes, telles que la protéinase K, la peptidase ou le lysozyme, sont ajoutées au surnageant pour rompre les liaisons peptidiques.
L’ADN est récupéré du surnageant par précipitation en ajoutant de l’alcool et un sel, comme l’acétate de sodium.
Le précipité isolé est dissous dans l’eau ou dans un tampon.

Les protocoles d'isolement de l'ADN peuvent être rapides et simples ou complexes et longs selon le type et la qualité de l'ADN requis pour un traitement ultérieur. Par exemple, l’extraction de l’ADN plasmidique est un peu plus compliquée que l’extraction de l’ADN génomique en raison de la nécessité d’une méthode de lyse appropriée pour séparer l’ADN plasmidique de l’ADNg pendant l’isolement. Cependant, pour des applications spécifiques, telles que le séquençage d’ADN à longue distance qui nécessite un bon rendement en échantillons d’ADN de haute qualité, nous devons suivre des protocoles spécifiques, même pour l’extraction d’ADN génomique.

Types de méthodes d'extraction d'ADN génomique

L’objectif principal des méthodes d’extraction de l’ADN génomique est de séparer l’ADNg des protéines, de l’ARN et des autres contenus cellulaires. Cela implique quatre étapes de base :

La plupart des protocoles d’isolement de l’ADN génomique sont des méthodes d’extraction en solution ou en phase solide. Les méthodes basées sur des solutions reposent sur des étapes de précipitation et de centrifugation pour séparer l'ADN des autres matériaux cellulaires, suivies d'une extraction organique ou d'un « relargage » pour séparer l'ADN soluble des protéines cellulaires. La précipitation finale de l'ADN se fait à l'aide d'éthanol. En revanche, les méthodes d'extraction en phase solide utilisent un support solide, tel que des matrices de silice ou de cellulose, pour lier l'ADN, suivi d'un lavage et d'une élution de l'ADN à partir du support solide. Cela implique des méthodes de centrifugation, de vide ou magnétiques pour séparer l’ADN lié des autres composants cellulaires.

Le choix de la méthode d’extraction de l’ADNg dépend du type d’échantillon, du nombre d’échantillons à traiter simultanément et de l’application de l’ADN en aval.

Genomic DNA extraction methods separate DNA from proteins, RNA, and other cell material. The process usually follows four main steps. First, the cell structure is disrupted with mechanical or chemical methods to make a cell lysate. Then the DNA is protected from damage during processing. After that, the soluble DNA is separated from cell debris. Finally, the purified DNA is eluted, or washed out, for later use.

DNA isolation can follow two main approaches: solution-based methods or solid-phase extraction methods. Solution-based methods use precipitation and centrifugation to separate DNA from other cellular material. They may also use organic extraction or salting out to remove proteins from the soluble DNA. The final DNA is then precipitated with ethanol.

Solid-phase extraction methods use a solid support, such as silica or cellulose matrices, to bind the DNA. The sample is then washed so other cellular components are removed. Next, the DNA is eluted from the solid support. These methods may use centrifugation, vacuum, or magnetic steps to separate the bound DNA during the process.

The best DNA extraction method depends on the sample type, how many samples must be processed, and the next step for the DNA. Plasmid DNA extraction is usually more complex than genomic DNA extraction because the lysis method must separate plasmid DNA from genomic DNA during isolation. In some cases, such as long-range DNA sequencing, specific protocols are needed to obtain enough high-quality DNA.
