FISH - Fluorescent In-situ Hybridization

L’hybridation in situ par fluorescence ou FISH est une technique utilisée pour détecter l’emplacement de gènes spécifiques ou de parties de gènes sur des chromosomes à l’intérieur de cellules fixes.
Dans cette technique, un ADN simple brin marqué par fluorescence est utilisé comme sonde pour lier des parties des chromosomes qui contiennent une séquence d’ADN complémentaire, appelée cible.
Tout d’abord, les cellules sont arrêtées en interphase ou en métaphase. Les cellules arrêtées sont suspendues dans une solution tampon et étalées sur une lame de verre propre.
Ensuite, l’ADN cible et les sondes marquées sont dénaturés par la chaleur ou des produits chimiques. Cette étape sépare l’ADN double brin, laissant à la fois la sonde et l’ADN cible sous forme de séquences simple brin capables d’appariement de bases.
Ensuite, la sonde et l’ADN cible sont mélangés et incubés pendant la nuit. Cela permet à la sonde de s’hybrider avec sa séquence complémentaire sur les chromosomes.
Une fois l’hybridation terminée, l’excès de sonde est emporté.
Si la sonde a été marquée avec un fluorophore, les emplacements de l’ADN hybride peuvent être directement visualisés avec un microscope fluorescent.
Si ce n’est pas le cas, l’ADN hybridé doit être marqué par fluorescence avant d’être visualisé. Pour ce faire, la séquence de la sonde doit contenir un nucléotide modifié lié à un haptène, une petite molécule à laquelle le fluorophore, tel qu’un anticorps fluorescent, peut se fixer.
Une telle visualisation directe des gènes est très utile pour les tests de diagnostic cellulaires, où des anomalies génétiques peuvent être détectées à partir de toute perte..., gain..., ou réarrangement des chromosomes pendant la division cellulaire.

L’hybridation in situ par fluorescence, ou FISH, a été développée au début des années 1980 et est rapidement devenue l’une des techniques les plus utilisées en cytogénétique. Les sondes marquées sont utilisées pour lier des séquences complémentaires d’ADN ou d’ARN sur un chromosome ou dans une région d’une cellule. Auparavant, les sondes ne pouvaient être obtenues que par clonage ou transcription inverse d’une matrice d’ADN. Actuellement, les oligonucléotides sondes peuvent être synthétisés par synthèse. De plus, grâce aux progrès des techniques optiques, des caméras CCD et des logiciels de traitement d'image sophistiqués, les signaux FISH peuvent être interprétés avec une plus grande sensibilité et une grande spécificité.

Ces avancées technologiques ont élargi la portée des applications FISH, de la cytogénétique et de la cartographie génétique au diagnostic des maladies génétiques et infectieuses. Les images haute résolution obtenues grâce à FISH aident à identifier les aberrations chromosomiques, telles que l'aneuploïdie commune (nombre anormal de chromosomes dans une cellule), les syndromes de microdélétion/microduplication (perte ou gain d'un chromosome) et les réarrangements subtélomériques.

FISH est également utilisé pour détecter la présence d’agents pathogènes microbiens dans les cellules sanguines humaines. Dans de tels tests de diagnostic, un court ADN marqué par fluorescence se lie à l’ARNr 16s microbien et peut être visualisé sous un microscope à fluorescence. Cela aide à l’identification des agents infectieux au niveau du genre ou de l’espèce. Les diagnostics basés sur FISH permettent une identification rapide de l'agent pathogène, ce qui est crucial pour détecter les organismes à croissance lente comme les mycobactéries, l'agent causal de la tuberculose, ou d'autres microbes, tels que Tropheryma whipplei, difficiles à cultiver.

FISH is a DNA and RNA detection method used in cytogenetics and medical testing. FISH stands for fluorescence in situ hybridization. It uses labeled probes that bind to complementary DNA or RNA sequences on a chromosome or inside a cell.

The method was developed in the early 1980s and became widely used because it can show specific genetic targets with high detail. In the past, probes were made by cloning or by reverse transcription of a DNA template. Today, probe oligonucleotides can also be synthesized synthetically.

Better optical tools have improved FISH results. CCD cameras and image-processing software help scientists read FISH signals with greater sensitivity and high specificity. These advances expanded FISH from gene mapping and cytogenetics into the diagnosis of genetic and infectious diseases.

FISH images can reveal chromosomal aberrations, or chromosome changes. These include common aneuploidy, which means an abnormal number of chromosomes in a cell, as well as microdeletion and microduplication syndromes, where a small chromosome region is lost or gained. FISH can also help identify subtelomeric rearrangements.

FISH is also used to detect microbial pathogens in human blood cells. In these tests, short fluorescence-labeled DNA binds to microbial 16s rRNA and can be seen with a fluorescence microscope. This helps identify infectious agents at the genus or species level.

Because FISH gives rapid results, it is useful for finding slow-growing organisms such as Mycobacteria, the bacteria that cause tuberculosis. It also helps detect microbes that are difficult to culture, such as Tropheryma whipplei. For this reason, FISH-based diagnostics are important in both genetics and infection testing.

EXPLANATION: The utilization of new technologies in any field of science is dependent on both economic and social factors. In addition to evaluating these factors, scientists must also consider long-term consequences that may not be initially apparent. The emphasis should be on the construction of a rationale argument that supports a position on the use of an application with ethical and social impact. Students should begin to appreciate the differences in ethical values that exist, and recognize that discussion of these values is imperative as knowledge and technology continue to advance, even when resolutions of differences can be rare. After investigation of a specific biotechnology application, students can write a position paper and/or participate in a classroom debate.
