Maxam-Gilbert Sequencing

Dans la méthode de Maxam-Gilbert, ou clivage chimique, l'ADN matrice est d'abord dénaturé et radiomarqué à l'extrémité 5' avec un phosphore 32. 
La réaction de séquençage est exécutée dans quatre tubes réactionnels différents avec quatre produits chimiques différents avec la capacité de modifier différents nucléotides dans l’ADN. 
Par exemple, l’acide formique n’attaquera que les purines, l’adénine ou la guanine, tandis que l’hydrazine n’attaquera que les pyrimidines - cytosine ou thymine. De même, les produits chimiques comme le sulfate de diméthyle n’attaquent qu’un seul nucléotide.
La pipéridine est ensuite utilisée pour cliver l’ADN aux bases modifiées, générant des fragments d’ADN radiomarqués de différentes longueurs.
Les quatre réactions sont ensuite exécutées en parallèle sur un gel et visualisées par autoradiographie. La séquence de l’ADN matrice peut ensuite être déchiffrée en lisant l’autoradiographie de bas en haut

La même année que la découverte de la méthode de séquençage Sanger, un autre groupe de scientifiques, Allan Maxam et Walter Gilbert, ont démontré leur méthode de clivage chimique pour le séquençage de l'ADN. La méthode Maxam-Gilbert repose sur l'utilisation de différents produits chimiques capables de cliver la séquence d'ADN à des sites spécifiques, la séparation des fragments d'ADN résultants de taille variable par électrophorèse et le déchiffrement de la séquence d'ADN à partir des bandes de gel résultantes.

Les défis de la méthode Maxam-Gilbert

La méthode Maxam-Gilbert ne peut être utilisée que pour résoudre généralement jusqu'à 400 pb en une seule analyse, ce qui est comparativement inférieur aux autres méthodes de séquençage traditionnelles telles que le séquençage de Sanger. De plus, la nécessité de quantités relativement importantes d'ADN matrice, associée à l'utilisation de radio-isotopes et de produits chimiques dangereux, rend la méthode Maxam-Gilbert moins favorable pour une utilisation de routine. Cependant, en raison de l’utilisation directe d’ADN purifié, la méthode Maxam-Gilbert est plus précise que le séquençage Sanger.

Applications actuelles de la méthode Maxam-Gilbert

Chemical cleavage DNA sequencing uses specific chemicals to cut DNA at known sites. Allan Maxam and Walter Gilbert described this method in the same year that the Sanger sequencing method was discovered. It reads the DNA sequence by breaking purified DNA into fragments and then studying the fragment pattern.

The method depends on chemicals that cleave DNA at specific bases. The resulting fragments are separated by electrophoresis, which is a way to sort DNA pieces by size using an electric field. After that, the sequence is determined from the band pattern seen on the gel.

The Maxam-Gilbert method has important limits. It usually resolves up to about 400 base pairs in one run, which is less than some traditional sequencing methods such as Sanger sequencing. It also needs relatively large amounts of template DNA, and it uses radioisotopes and hazardous chemicals, so it is not ideal for routine use.

Even with these drawbacks, the method has one advantage. Because it works directly with purified DNA, it can be more accurate than Sanger sequencing. Today, it is used less often, but it still has value in some DNA fingerprinting methods and in DNA structural studies.

Students may also consider the pros and cons of biotechnical applications. Investigation of techniques utilized could come from field trips to witness the use of these techniques in practice, lab exercises, virtual labs, simulation activities, interviews with professionals, etc. Molecular techniques could include: PCR, electrophoresis, restriction enzyme digestion of DNA, DNA sequencing, plasmid-based transformation, transfection, etc. These techniques are used in fields of medicine, agriculture, biomedical engineering, forensic science, etc.
