La cryomicroscopie électronique ou cryo-EM examine des échantillons biologiques congelés à des températures cryogéniques inférieures à −150 °C.
Contrairement aux EM conventionnelles, la cryo-EM ne nécessite pas de fixer et de déshydrater un échantillon, ce qui aide à préserver l’état natif des structures biologiques.
Les électrons de haute énergie utilisés dans les EM peuvent endommager les liaisons chimiques d’un échantillon. La congélation de l’échantillon permet d’éviter ces dommages, ce qui permet d’obtenir des images à une résolution proche de l’atome.
Dans la cryo-EM, de petits échantillons biologiques en solution sont vitrifiés, c’est-à-dire rapidement congelés, dans de l’éthane liquide. La vitrification fixe l’échantillon dans son état natif dans une fine couche d’eau, et l’eau dans l’échantillon forme un état semblable à du verre sans l’apparition de cristaux de glace.
Les échantillons plus épais, tels que les cellules et les tissus, ne peuvent pas être vitrifiés dans une fine couche liquide. Par conséquent, ces échantillons peuvent être étudiés à l’aide de la méthode cryo-EM des coupes vitrées. Dans cette méthode, un échantillon est d’abord vitrifié par congélation à haute pression, puis coupé en sections ultraminces à -140 °C avant d’être observé à l’aide de la cryo-EM.

Cryo-electron Microscopy

La microscopie électronique (ME) conventionnelle implique la déshydratation, la fixation et la coloration d'échantillons biologiques, ce qui déforme l'état natif des molécules biologiques et entraîne plusieurs artefacts. De plus, le faisceau d’électrons à haute énergie endommage l’échantillon et rend difficile l’obtention d’images à haute résolution. Ces problèmes peuvent être résolus grâce à la cryo-ME, qui utilise des échantillons congelés et des faisceaux d’électrons plus doux. La technique a été développée par Jacques Dubochet, Joachim Frank et Richard Henderson, pour laquelle ils ont remporté le prix Nobel de chimie en 2017.

Les techniques de diffraction des rayons X et de résonance magnétique nucléaire (RMN) sont également utilisées pour obtenir des structures à haute résolution de biomolécules ou étudier leurs changements conformationnels. Cependant, pour obtenir la structure des rayons X, une molécule doit être cristallisée, ce qui n’est pas toujours possible. Même si une molécule est cristallisable, le processus de cristallisation peut altérer sa structure biomoléculaire, qui n'est plus représentative de la structure native. Cryo-EM ne nécessite pas d'échantillon cristallisé et permet également de visualiser les mouvements moléculaires ou les interactions de biomolécules dans leur état natif. De même, la technique RMN n’est limitée qu’à des protéines solubles relativement petites et ne convient pas aux protéines non polaires intégrées dans les membranes cellulaires. D'autre part, la cryo-ME permet l'étude de protéines plus grosses, de récepteurs liés à la membrane ou de complexes biomoléculaires.

Cryo-ME est largement utilisé en biochimie pour étudier la structure, la fonction et l'interaction des biomolécules. Les agents infectieux, tels que les virus, peuvent être identifiés et étudiés grâce à la cryo-ME. Par exemple, lors de l’épidémie du virus Zika au Brésil, la cryo-ME a été utilisée pour créer une image 3D de la structure du virus afin que les cibles potentielles des médicaments antiviraux puissent être identifiées.

Cryo-EM, or cryo-electron microscopy, lets scientists image biomolecules in their frozen, native state. It avoids the dehydration, fixation, and staining used in conventional electron microscopy, which can distort samples and create artifacts. It also reduces damage from the electron beam by using gentler imaging conditions.

This method is useful when other structure tools have limits. X-ray diffraction needs a molecule to form a crystal, and that is not always possible. Even when crystallization works, it can change the biomolecule’s structure so it no longer matches the native form.

Cryo-EM also fills gaps left by nuclear magnetic resonance, or NMR. NMR is mainly used for relatively small soluble proteins and is not suited to non-polar proteins in cell membranes. Cryo-EM can be used to study larger proteins, membrane-bound receptors, and biomolecular complexes.

Researchers use cryo-EM to study biomolecule structure, function, and interactions. It can also help identify and analyze infectious agents such as viruses. During the Zika virus outbreak in Brazil, cryo-EM was used to build a 3D image of the virus so potential antiviral drug targets could be found. The technique was developed by Jacques Dubochet, Joachim Frank, and Richard Henderson, who shared the 2017 Nobel Prize in Chemistry.
