Human Proximal Airway

The human proximal airway comprises the central conducting passages, including the trachea and main bronchi, that transport air toward the lungs while protecting deeper respiratory tissues. Its epithelial lining supports mucociliary clearance: mucus captures particles and pathogens, and coordinated ciliary beating moves this material toward the throat for removal. In bioengineering, these structural and functional features guide the development of airway-on-a-chip systems, tissue-engineered grafts, and in vitro models that reproduce airway barriers, airflow, and host responses. Such platforms support respiratory disease research, drug testing, biomaterial evaluation, and future strategies for airway repair and regeneration.

This paper describes different methods of growing Pseudomonas aeruginosa biofilms on cultured human airway epithelial cells. These protocols can be adapted to study different aspects of biofilm formation, including visualization of the biofilm, staining of the biofilm, measuring the colony forming units (CFU) of the biofilm, and studying biofilm cytotoxicity.
