Intracellular Calcium Signaling

Intracellular calcium signaling is the regulated movement and interpretation of calcium ions within cells, enabling rapid communication that coordinates essential biological functions. Stimuli activate calcium entry through plasma-membrane channels or release from intracellular stores such as the endoplasmic reticulum, producing localized or cell-wide calcium changes that bind sensor proteins and alter enzyme activity, gene expression, contraction, or secretion. In bioengineering, researchers measure and manipulate these signals to study cell behavior, design responsive biomaterials, and develop engineered tissues and therapeutic systems. Calcium imaging and optogenetic or chemical control of calcium dynamics also help link signal patterns to specific cellular outcomes.

Monitoring Changes in the Intracellular Calcium Concentration and Synaptic Efficacy in the Mollusc Aplysia

We demonstrate how changes in the intracellular free calcium concentration and synaptic efficacy can be simultaneously monitored in a ganglion preparation of Aplysia. We image intracellular calcium using a fluorescent dye, Calcium Orange, and induce and monitor synaptic transmission with sharp (intracellular) electrodes.
