Molecular Diagnostics

Molecular diagnostics is the detection and analysis of DNA, RNA, proteins, or other molecular markers to identify disease, characterize biological states, or guide treatment. These methods use processes such as nucleic acid extraction, sequence-specific hybridization, polymerase chain reaction amplification, sequencing, or biomolecular binding to convert molecular signals into measurable results. In bioengineering, molecular diagnostics supports the design of biosensors, microfluidic platforms, and integrated assays for detecting pathogens, genetic variants, and cancer-associated biomarkers. By improving sensitivity, specificity, speed, and portability, these technologies enable earlier diagnosis, personalized medicine, environmental monitoring, and research on disease mechanisms.

We present a complete protocol for postmortem diagnosis of animal rabies under field conditions using a rapid immunochromatographic diagnostic test (RIDT), from brain biopsy sampling to final interpretation. We also describe further applications using the device for molecular analysis and viral genotyping.

Here we report the generation of Tre recombinase through directed, molecular evolution. Tre recombinase recognizes a pre-defined target sequence within the LTR sequences of the HIV-1 provirus, resulting in the excision and eradication of the provirus from infected human cells. While still in its infancy, directed molecular evolution will allow the creation of custom enzymes that will serve as tools of molecular surgery and molecular medicine.
