Recellularization Protocol

A recellularization protocol is a structured method for repopulating a decellularized tissue or organ scaffold with living cells, helping restore biological function while preserving the native extracellular matrix architecture. Typically, the protocol introduces selected cells into the scaffold under controlled conditions, often using perfusion to distribute cells through the vascular network and support their adhesion, proliferation, and differentiation. In bioengineering, recellularization supports the development of engineered tissues and organ models for studying disease, testing therapies, and advancing regenerative medicine. Its success depends on scaffold integrity, cell compatibility, culture conditions, and effective integration between newly introduced cells and the remaining matrix.

Perfusion decellularization is a novel technique to produce whole liver scaffolds that retains the organ's extracellular matrix composition and microarchitecture. Herein, the method of preparing whole organ scaffolds using perfusion decellularization and subsequent repopulation with hepatocytes is described. Functional and transplantable liver grafts can be generated using this technique.

The description of a nonhuman primate model of lung regeneration in which whole lungs are decellularized and then seeded with adult stem cells and endothelial cells in a bioreactor that facilitates vascular circulation and liquid media ventilation is presented.

Neutrophils are among the first cells to arrive on the site of inflammatory immune response, and their functions and mechanisms have been studied extensively in vitro. We demonstrate a standard density gradient separation method to isolate human neutrophils from whole blood using commercially available separation media.
