Reporter Protein Expression

Reporter protein expression is the production of measurable proteins that reveal when, where, or how strongly a gene or regulatory sequence is active. In bioengineering, researchers place a reporter gene under a target promoter or fuse it to a protein of interest, so cellular transcription and translation generate a detectable signal such as fluorescence or luminescence. Measuring this signal enables analysis of promoter activity, gene regulation, protein localization, and changes in cellular state. Reporter systems support the design and optimization of engineered genetic circuits, evaluation of delivery strategies, and characterization of biological processes in research and biotechnology.

Generation of recombinant rotaviruses from plasmid DNA provides an essential tool for the study of rotavirus replication and pathogenesis, and the development of rotavirus expression vectors and vaccines. Herein, we describe a simplified reverse genetics approach for generating recombinant rotaviruses, including strains expressing fluorescent reporter proteins.

Circadian clocks function within individual cells, i.e., they are cell-autonomous. Here, we describe methods for generating cell-autonomous clock models using non-invasive, luciferase-based real-time bioluminescence technology. Reporter cells provide tractable, functional model systems for studying circadian biology.
