Vascularized Skin Equivalents

Vascularized skin equivalents are engineered three-dimensional skin substitutes that reproduce both the layered structure of skin and its blood vessel network, addressing a major limitation of conventional skin models and grafts. They typically combine keratinocytes, fibroblasts, and vascular cells within a biomaterial scaffold, where endothelial cells organize into microvessels that can support nutrient and oxygen transport through perfusion. In bioengineering, these constructs provide more physiologically relevant platforms for studying wound healing, vascularization, skin disease, and drug responses. They may also improve the integration and survival of grafts used for burns, chronic wounds, and tissue reconstruction, advancing regenerative medicine and personalized testing.

The goal of this protocol is to build up a three-dimensional full thickness skin equivalent, which resembles natural skin. With a specifically constructed automated wounding device, precise and reproducible wounds can be generated under maintenance of sterility.

They must adhere to compendial or applicable standards for potency, content uniformity, disintegration times, and dissolution rates.
