Von Kossa Assay

The Von Kossa assay is a histochemical method for detecting mineral deposition, particularly phosphate-containing calcium salts, in cells, tissues, and engineered biomaterials. In the procedure, silver ions replace phosphate ions within mineral deposits; exposure to light reduces the bound silver to metallic silver, producing dark brown or black staining at mineralized sites. In bioengineering, this assay helps evaluate osteogenic differentiation, bone formation, and the mineralization of scaffolds or tissue-engineered constructs. Staining patterns provide a qualitative measure of matrix mineralization and support the assessment of biomaterial performance and bone-regenerative strategies.
