Axon Guiding Microchannels

Axon guiding microchannels are engineered, microscale pathways that direct the growth of neuronal axons, helping organize connections in controlled laboratory or biomedical settings. Their narrow, typically parallel channels provide physical confinement and topographical cues that encourage axons to extend along defined paths while limiting lateral growth, often across a gap or between neuronal populations. These platforms support studies of axon development, neuronal connectivity, and injury-related regeneration, and can help evaluate how neurons respond to biochemical or mechanical conditions. In neural tissue engineering, microchannels also provide a framework for designing organized neural interfaces and guiding reconnection after damage.

A microchannels-on-a-chip platform was developed by the combination of photolithographic reflowable photoresist technique, soft lithography, and microfluidics. The endothelialized microchannels platform mimics the three-dimensional (3D) geometry of in vivo microvessels, runs under controlled continuous perfusion flow, allows for high-quality and real-time imaging and can be applied for microvascular research.

Source: Li, Q., et al. Investigating Mammalian Axon Regeneration: In Vivo Electroporation of Adult Mouse Dorsal Root Ganglion. J. Vis. Exp. (2018)This video demonstrates the process of axonal regeneration in a mouse sciatic nerve following a crush injury. The isolated nerve is fixed and prepared for imaging, with fluorescent proteins expressed in neurons to visualize axon growth. Using an epifluorescence microscope, axon regeneration is traced from the injury site to assess the extent of...
