Culturomics

Culturomics is a high-throughput microbiological approach that systematically cultivates and identifies microorganisms under many laboratory conditions, expanding access to microbial diversity beyond standard culture methods. By varying nutrient media, oxygen levels, temperature, incubation times, and other growth parameters, researchers create complementary conditions that favor different organisms, then characterize isolates with tools such as mass spectrometry or genetic sequencing. In biology, this strategy can recover previously unrecognized or difficult-to-culture bacteria, reveal community composition, and support studies of microbial physiology, ecology, and host-microbe interactions. Culturomics also provides strains for functional experiments, diagnostics, biotechnology, and microbiome research.

This protocol enables rapid, cost-effective evaluation of microbial composition in dairy products by combining culturomics with MALDI-TOF MS. It delivers taxonomy results comparable to 16S rRNA sequencing with a shorter turnaround time than traditional culture methods.

Isolation of Tomato Seed-Associated Microbiota Through Controlled Germination and Comparative Culturing

2026

This protocol combines standardized seed surface sterilization with controlled germination to enable the reproducible isolation of tomato seed-associated microbiota during the seed-to-seedling transition. The approach enhances recovery of culturable microorganisms that are underrepresented in dry seeds and supports comparative culturomic studies during early seedling development.
