Feature Counts

Feature counts are numerical tallies of biological features, such as genes or transcripts, detected in high-throughput sequencing data and used to convert raw reads into interpretable measurements. In RNA sequencing, computational pipelines align or map reads to a reference genome or transcriptome, assign them to annotated features according to defined overlap and counting rules, and organize the results in a count matrix. Researchers use these matrices to compare gene expression across samples, identify differentially expressed features, classify cell types, and investigate biological responses. Reliable counting supports reproducible genomic analysis and provides a foundation for downstream statistical modeling and systems biology.

Using a Hemacytometer to Count Cells

2023

Many biomedical experiments require manipulation of a known quantity of cells, in order to achieve accurate, reproducible, and statistically-relevant data. Therefore, learning how to count cells is a particularly essential technique for any successful biomedical scientist. The most common way to count cells is by using a hemacytometer - an instrument that bears two laser-etched grids, which aid in the enumeration of an aliquot cells under a simple light microscope. This data can then be used to...

Knowledge of the exact number of viable cells is required for many tissue culture manipulations. This protocol describes how to differentiate between live and dead cells and quantify cells using a hemacytometer. Although it describes counting human neural stem/precursor cells (hNSPCs), it can be used for other cell types.

Visual Search for Features and Conjunctions

Source: Laboratory of Jonathan Flombaum—Johns Hopkins University How do people find objects in cluttered visual scenes? Think, for example, of looking for keys on a messy desk, finding the ripest-looking fruit at the grocery store, locating your car when you can’t quite remember where you parked it, or finding an old friend at an airport exit gate. Clearly, an understanding of visual perception is going to play a role in any answers, and more specifically, an understanding of visual attention...
