Thrombin

Thrombin is a serine protease that regulates blood coagulation by converting soluble fibrinogen into an insoluble fibrin mesh, making it central to hemostasis and wound repair. Generated from prothrombin within the coagulation cascade, thrombin cleaves fibrinogen and activates platelets and clotting factors V, VIII, XI, and XIII, amplifying and stabilizing clot formation. Studying thrombin helps explain how bleeding is controlled and how excessive coagulation contributes to thrombosis. Its activity is measured in coagulation assays and targeted by anticoagulant drugs, while purified thrombin is also used in clinical hemostatic products and biomaterial research.

The Nijmegen Hemostasis Assay: Simultaneous Fluorogenic Measurement of Thrombin and Plasmin Generation in a Single Well

2026

This protocol describes the Nijmegen Hemostasis Assay, which enables simultaneous, time-resolved measurement of thrombin and plasmin generation to provide an integrated assessment of coagulation and fibrinolysis. The assay aims to improve characterization of hemostatic balance in research and clinical settings beyond the scope of conventional hemostatic tests.

RNA-seq Analysis of Transcriptomes in Thrombin-treated and Control Human Pulmonary Microvascular Endothelial Cells

This protocol presents a complete and detailed procedure to apply RNA-seq, a powerful next-generation DNA sequencing technology, to profile transcriptomes in human pulmonary microvascular endothelial cells with or without thrombin treatment. This protocol is generalizable to various cells or tissues affected by different reagents or disease states.

Purification and Activation of Platelets from Murine Whole Blood

2025

In this video, we describe a protocol for purifying platelets from murine whole blood and their subsequent activation with the platelet agonist thrombin. When analyzed with flow cytometry, the purified resting platelets are characterized based on the expression of their cell-surface marker CD41, whereas purified activated platelets co-express both the surface markers CD41 and CD62P.

This study describes a novel microplate assay that measures FV coagulation activity during fibrin clot formation in human plasma which has not been reported previously. The method uses a kinetic microplate reader to continuously measure the change in absorbance at 405nm during fibrin clot formation in human plasma.
