Primary Tumor Growth

Primary tumor growth is the progressive increase in size and cellular mass of a tumor at its original site, a central process in cancer development and research. It occurs when genetic and epigenetic changes disrupt controls on cell proliferation, survival, and tissue organization, while tumor cells recruit blood vessels through angiogenesis and interact with the surrounding microenvironment. Measuring growth kinetics, cellular heterogeneity, and responses to treatment helps researchers characterize tumor biology and identify therapeutic vulnerabilities. Studying primary tumors also clarifies how cancers adapt before invasion or metastasis, supporting improved models for diagnosis, drug testing, and treatment design.

The Influence of Liver Resection on Intrahepatic Tumor Growth

A high incidence of tumor recurrence after resection of liver metastases remains an unsolved problem. The illustrated mouse model may be useful to investigate the reasons for such recurrences. It combines a liver resection model with intrahepatic tumor cell injection for the first time.

A modified 3-D in vitro system is presented in which growth characteristics of several tumor cell lines in reconstituted basement membrane correlate with the dormant or proliferative behavior of the tumor cells at a metastatic secondary site in vivo.

Primary Tumor and MEF Cell Isolation to Study Lung Metastasis

The goal of this protocol is to study breast tumorigenesis. With this technique, mouse mammary tumors are removed and primary cells are prepared from tumors. A lung extraction protocol is included for studying lung metastasis. Furthermore, another protocol for analyzing mouse embryonic fibroblasts from the mouse embryo is included.

In this report, we describe a protocol for isolating highly purified populations of leukocytes that infiltrate tumors. This protocol is adapted from the Miltenyi Biotech protocol to enhance yield and purity for isolating cells from complex tumor tissue.
