Internal Standard Calibration

Internal standard calibration is a quantitative analytical method that improves measurement accuracy by comparing an analyte’s signal with the signal from a known internal standard. In practice, analysts add a fixed amount of a chemically similar compound to calibration solutions and samples, then construct a calibration curve from the analyte-to-standard response ratio; this ratio compensates for variations in sample preparation, injection volume, instrument response, or signal loss. The approach supports reliable concentration measurements in techniques such as gas chromatography, liquid chromatography, and mass spectrometry, especially when matrix effects or instrumental drift can affect raw signals.
