Maldi Plate Loading

MALDI plate loading is the preparation of samples and matrix on a target plate for matrix-assisted laser desorption/ionization mass spectrometry, where careful deposition strongly influences data quality. Typically, an analyte is mixed with an energy-absorbing matrix and a small volume is applied to a designated spot, then dried to form co-crystals that support efficient laser-driven desorption and ionization. Consistent spotting, appropriate sample-to-matrix ratios, and thorough drying help produce uniform crystals, stronger signals, and improved mass accuracy. In chemistry, reliable MALDI plate loading supports the analysis of peptides, proteins, polymers, and other molecules, enabling rapid molecular-weight determination and structural characterization.

MALDI-TOF Mass Spectrometry

2023

Matrix-assisted laser desorption ionization (MALDI) is a mass spectrometry ion source ideal for the analysis of biomolecules. Instead of ionizing compounds in the gaseous state, samples are embedded in a matrix, which is struck by a laser. The matrix absorbs the majority of the energy; some of this energy is then transferred to the sample, which ionizes as a result. Sample ions can then be identified using a time-of-flight analyzer (TOF). This video covers principles of MALDI-TOF, including...
