Microdroplet Chips

Microdroplet chips are microfluidic devices that generate, transport, and manipulate tiny, isolated droplets, creating controlled reaction environments for chemical analysis and synthesis. Within their channels, immiscible fluids are segmented into droplets by flow conditions and channel geometry; each droplet acts as a miniature reactor whose composition, residence time, and interfacial area can be regulated. This compartmentalization reduces sample consumption and cross-contamination while supporting rapid mixing, heat transfer, and mass transfer. In chemistry, microdroplet chips enable high-throughput reaction screening, particle and material formation, analytical assays, and studies of reaction kinetics with improved control and reproducibility.

Source: Jian, X., et al., Automated Microbial Cultivation and Adaptive Evolution using Microbial Microdroplet Culture System (MMC). J. Vis. Exp. (2022).This video demonstrates the use of a microbial microdroplet culture system to drive adaptive evolution of bacteria toward methanol tolerance. By mixing engineered bacterial cultures with a high-methanol stress medium and segmenting them into microdroplets, the system selects for mutants that exhibit improved growth under methanol stress. Here, we demonstrate a simple production method for size-controllable, monodisperse, water-in-oil (W/O) microdroplets using a capillary-based centrifugal microfluidic device. This method requires only a small sample volume and enables high-yield production. We expect this method will be useful for rapid biochemical and cellular analyses.

Chip-Based Digital PCR to Detect Rare Transcript Variants Using a Nanofluidic Chip

2025

This video demonstrates chip-based digital PCR — a variation of the digital PCR technique that is useful in detecting rare transcript variants. The PCR reaction is partitioned into the chambers of a nanofluidic chip, each of which acts as an independent reaction. The detection of fluorescence signals from the chambers with amplified targets confirms the presence of rare transcript variants in the sample.

Introduction to Organ-on-Chip Models Organ-on-chip models are advanced microphysiological systems that recreate key structural and functional features of human organs in a controlled laboratory setting. These devices integrate living human cells within microfluidic channels, allowing researchers to simulate tissue–tissue interfaces, fluid flow, and mechanical forces that are essential for normal organ function. Unlike traditional cell cultures, which often lack these dynamic physiological cues,...
