Transwell Permeability

Transwell permeability is an in vitro method for measuring how readily a dissolved compound crosses a porous membrane or cultured cell layer, providing a controlled way to assess transport and barrier function. In a typical insert, the test solution is placed in a donor compartment above the membrane, and samples are collected from the receiver compartment over time; concentration changes yield permeability metrics such as apparent permeability (Papp). Membrane properties, concentration gradients, temperature, and cellular junctions can influence the result. In chemistry and pharmaceutical research, the assay supports screening molecular transport, evaluating formulation behavior, and comparing passive diffusion with cell-mediated barrier effects.

We describe a live whole animal quantitative measurement for permeability of the embryonic zebrafish brain. The technique analyzes the ability to retain cerebrospinal fluid and molecules of different molecular weights within the neural tube lumen and quantifies their movement out of the ventricles. This method is useful for determining differences in epithelial permeability and maturation during development and disease.
