Pancreatic Cancer

Pancreatic cancer is a group of malignant diseases that arise in the pancreas, an organ involved in digestion and blood-glucose regulation, and it remains a major clinical challenge because early disease may produce few noticeable symptoms. Cancer develops when genetic and cellular changes disrupt normal control of cell growth, allowing abnormal cells to survive, invade nearby tissues, and potentially spread to distant organs. Clinical evaluation integrates symptoms, imaging, laboratory findings, biopsy, and disease staging to guide management. Treatment may include surgery, chemotherapy, radiation therapy, targeted approaches, or combinations of these, while ongoing research seeks earlier detection and more effective personalized therapies.

Isolation of Stem Cells from Human Pancreatic Cancer Xenografts

Cancer stem cells (CSCs) have been identified in a number of malignancies. In this protocol we describe a flow cytometric method utilizing aldehyde dehydrogenase activity and CD44 and CD24 expression to isolate CSCs from human pancreatic adenocarcinoma xenografts. These viable cells can then be used in functional and analytical studies.

Bioluminescent Orthotopic Model of Pancreatic Cancer Progression

Improved understanding of pancreatic cancer biology is critically needed to enable the development of better therapeutic options to treat pancreatic cancer. To address this need, we demonstrate an orthotopic model of pancreatic cancer that permits non-invasive monitoring of cancer progression using in vivo bioluminescence imaging.

Pancreatic cancer stem cells (CSCs) can be expanded in vitro using the anchorage-independent sphere culture technique, which represents a powerful tool to study CSC biology and can serve as the first step to develop novel CSC-targeting therapies. Here the methodology for expanding, analyzing and targeting of pancreatic CSCs is provided.
