Thyroid Nodules

Thyroid nodules are discrete growths or fluid-filled lesions within the thyroid gland that matter because they may alter hormone production or, less commonly, represent thyroid cancer. They arise through localized proliferation of thyroid cells, cyst formation, or other structural changes and are evaluated using medical history, thyroid-stimulating hormone testing, ultrasound features, and, when indicated, fine-needle aspiration biopsy. Clinical assessment distinguishes benign nodules from those requiring surveillance or treatment, including surgery or targeted therapy. Understanding nodule structure and function supports accurate risk stratification, avoids unnecessary procedures, and guides personalized care for patients with thyroid disease.

This protocol describes dissection of mouse embryonic thyroid anlagen and the culture of explants on semiporous filters or on microscopy plastic slides. This system is ideal to study morphogenetic or differentiation events occurring during thyroid development of wild type or knockout embryos, and is amenable to gain- and loss-of-function experiments.
