Epidermal Progenitor Cells

Epidermal progenitor cells are tissue-resident precursor cells that replenish the epidermis, the continuously renewing outer layer of skin, and provide a model for studying tissue development and maintenance. They preserve the basal cell population through self-renewal while producing daughter cells that progressively differentiate into keratinocytes and migrate toward the skin surface as they form the protective barrier. In developmental biology, their regulation helps explain epidermal stratification, wound repair, and responses to changing tissue demands; understanding these cells also supports research on inherited skin disorders, regenerative medicine, and engineered skin substitutes.

Here we show our method to isolate mouse epidermal neural crest stem cells (EPI-NCSC). Technique involves micro-dissecting whisker follicles, isolating the bulge and placeing it into tissue culture. EPI-NCSC start to emigrate from bulge explants onto the substratum within 3 - 4 days.
