Epithelial Cell Types

Epithelial cell types are specialized cells that form continuous, polarized sheets lining body surfaces, cavities, and developing organs, where they create barriers and regulate exchange with underlying tissues. Their structure and function depend on apical-basal polarity, selective cell junctions, and coordinated changes in adhesion, shape, proliferation, and differentiation. During development, these properties enable epithelial folding, branching, tubule formation, and transitions to migratory cell states that shape tissues and organs. Identifying distinct epithelial populations helps researchers trace lineage relationships, study organ morphogenesis, and understand how disrupted differentiation or barrier function contributes to congenital abnormalities and disease.

This protocol presents a flow cytometry-based method to isolate viable alveolar type II epithelial cells (AECII) from neonatal, juvenile, or adult murine lungs (infected or non-infected). This approach enables downstream molecular and/or functional studies of AECII to identify their roles during lung inflammation and infection.

Flow Cytometric Isolation of Primary Murine Type II Alveolar Epithelial Cells for Functional and Molecular Studies

We describe the rapid isolation of primary murine type II alveolar epithelial cells (AECII) by flow cytometric negative selection. These AECII show high viability and purity and are suitable for a wide range of functional and molecular studies regarding their role in respiratory conditions such as autoimmune or infectious diseases.

We present a protocol that combines cell isolation and whole-cell patch-clamp recording to measure the electrical properties of the primary dissociated epithelial cells from the rat cauda epididymides. This protocol allows for investigation of the functional properties of primary epididymal epithelial cells to further elucidate the physiological role of the epididymis.
