Gtp Wash Specificity

GTP wash specificity describes the ability of a wash step to preserve interactions dependent on guanosine triphosphate (GTP) while removing nonspecific or weakly associated molecules. It relies on controlled wash conditions that distinguish GTP-bound complexes from unbound material, with factors such as nucleotide state, binding affinity, buffer composition, and wash stringency determining the outcome. In genetics and molecular biology, assessing GTP wash specificity helps evaluate nucleotide-dependent protein interactions and improve the reliability of binding, purification, and biochemical assays. Clear specificity reduces background signal, supports accurate interpretation of experimental results, and strengthens studies of regulatory pathways involving GTP-binding proteins.

Guanosine triphosphate (GTP) binding is one of the earliest events in G-Protein-Coupled Receptor (GPCR) activation. This protocol describes how to pharmacologically characterize specific GPCR-ligand interactions by monitoring the binding of the radio-labeled GTP analog, [35S]guanosine-5'-O-(3-thio)triphosphate ([35S]GTPγS), in response to a ligand of interest.

Washing, Drying, and Ignition of Precipitates

After filtration, the precipitate is washed to remove coprecipitated impurities and any remaining mother liquor. Colloidal precipitates, such as silver chloride, are washed with an electrolyte (such as dilute nitric acid) to prevent the peptization of the precipitate. In the case of slightly soluble precipitates, the wash solution contains a common ion to reduce solubility. Lead sulfate, which is slightly soluble in water, is washed with dilute sulfuric acid. Similarly, wash solutions may be...

Platelet metabolism is of interest, particularly as it relates to the role of platelet hyper- and hypoactivity in bleeding and thrombotic disorders. Isolating platelets from plasma is necessary for some metabolic assays; presented here is a method for isolating of intracellular metabolites from washed platelets.

Detection of Small GTPase Prenylation and GTP Binding Using Membrane Fractionation and GTPase-linked Immunosorbent Assay

Here we describe a protocol to investigate the prenylation and guanosine-5'-triphosphate (GTP)-loading of Rho GTPase. This protocol consists of two detailed methods, namely membrane fractionation and a GTPase-linked immunosorbent assay. The protocol can be used for measuring the prenylation and GTP loading of different other small GTPases.
