Dna Fiber Analysis

DNA fiber analysis is a molecular technique that visualizes and measures the movement of individual DNA replication forks, providing direct insight into genome duplication and stability. Cells are sequentially labeled with different thymidine analogs, such as CldU and IdU, before DNA is gently extracted, stretched onto microscope slides, and detected with fluorescent antibodies; the resulting tracks reveal replication-fork progression, origin spacing, fork asymmetry, and stalled forks. In genetics research, this method helps characterize replication dynamics, assess cellular responses to replication stress, and identify defects associated with genome instability and disease. It also supports evaluation of factors that regulate DNA replication and repair.

DNA Methylation: Bisulphite Modification and Analysis

The gold standard for DNA methylation analysis is genomic sequencing of bisulphite converted DNA. This method takes advantage of the increased sensitivity of cytosine compared with 5-methylcytosine (5-MeC) to bisulphite deamination under acidic conditions. Unmethylated cytosines can be distinguished from methylated cytosines after PCR amplification of the target genomic DNA.
