Live Larvae Counting

Live larvae counting is a quantitative method for determining the number of living larval organisms in a sample, providing a direct measure of survival, development, and population size. Researchers examine cultures or experimental samples under a microscope, distinguish live larvae from nonviable individuals by movement and morphology, and record totals across defined time points or conditions. In genetics, these measurements help evaluate how mutations, gene knockdown, environmental factors, or treatments affect viability, developmental progression, and phenotype frequency. Repeated counts can also support genetic screens, compare experimental groups, and reveal developmental defects that may not be apparent from endpoint measurements alone.

Drosophila larvae are an attractive model system for live imaging due to their translucent cuticle and powerful genetics. This protocol describes how to utilize a single-layer PDMS device, called the 'larva chip' for live imaging of cellular processes within neurons of 3rd instar Drosophila larvae.

Using a Hemacytometer to Count Cells

2023

Many biomedical experiments require manipulation of a known quantity of cells, in order to achieve accurate, reproducible, and statistically-relevant data. Therefore, learning how to count cells is a particularly essential technique for any successful biomedical scientist. The most common way to count cells is by using a hemacytometer - an instrument that bears two laser-etched grids, which aid in the enumeration of an aliquot cells under a simple light microscope. This data can then be used to...

Knowledge of the exact number of viable cells is required for many tissue culture manipulations. This protocol describes how to differentiate between live and dead cells and quantify cells using a hemacytometer. Although it describes counting human neural stem/precursor cells (hNSPCs), it can be used for other cell types.

Determining the number of cells in culture is important in standardization of culture conditions and in performing accurate quantitation experiments. In this video, we demonstrate how cells are counted using a hemacytometer.

A Technique to Establish a Bacterial Infection Model in Insect Larvae

2025

This video demonstrates a method to establish a Mycobacterium bovis infection model in the larvae of the greater wax moth, Galleria mellonella. A bioluminescent strain of Mycobacterium bovis BCG is injected into the hemolymph of the larva to establish infection, and the bioluminescence is measured at predefined intervals to study the progress of infection.
