Mrna Level Measurement

mRNA level measurement is the quantification of messenger RNA molecules in a biological sample, providing a snapshot of gene expression and regulation. In common workflows, RNA is isolated, converted into complementary DNA by reverse transcription, and quantified using real-time PCR, sequencing, or related hybridization-based methods; normalization to reference transcripts helps account for sample-to-sample variation. These measurements allow researchers to compare transcriptional responses across tissues, developmental stages, genetic backgrounds, or experimental conditions. In genetics, mRNA profiling connects DNA sequence and regulatory mechanisms with functional gene activity, supporting studies of disease-associated variants, cellular pathways, and responses to treatment.

Here, we present a protocol using RNA-seq to monitor mRNA levels over time during the hypoxic response of S. cerevisiae cells. This method can be adapted to analyze gene expression during any cellular response.

Measuring the Kinetics of mRNA Transcription in Single Living Cells

RNA polymerase II transcriptional kinetics are measured on specific genes in living cells. mRNAs transcribed from the gene of interest are fluorescently tagged and using Fluorescence Recovery After Photobleaching (FRAP) the in vivo kinetics of transcriptional elongation are obtained.

Measurement of mRNA Decay Rates in Saccharomyces cerevisiae Using rpb1-1 Strains

The steady state level of specific mRNAs is determined by the rate of synthesis and decay of the mRNA. Genome-wide mRNA degradation rates or the decay rates of specific mRNAs can be measured by determining mRNA half-lives. This protocol focuses on measurement of mRNA decay rates in Saccharomyces cerevisiae.

Ratio Level of Measurement

2023

The way a set of data is measured is called its level of measurement. Correct statistical procedures depend on a researcher being familiar with levels of measurement. For analysis, data are classified into four levels of measurement—nominal, ordinal, interval, and ratio. A set of data measured using the ratio scale takes care of the ratio problem and provides complete information. Ratio scale data are like interval scale data, except they have a zero point and ratios can be calculated. For...

The way a set of data is measured is called its level of measurement. Correct statistical procedures depend on a researcher being familiar with levels of measurement. For analysis, data are classified into four levels of measurement—nominal, ordinal, interval, and ratio. Data measured using an ordinal scale are similar to nominal scale data, but there is one major difference. The ordinal scale data can be ordered. An example of ordinal scale data is a list of the top five national parks in the...
