Mutant Isolation Protocol

A mutant isolation protocol is a systematic genetic method for identifying and recovering organisms or cells that carry heritable alterations in their DNA. It typically combines mutagenesis or the use of existing variant populations with selection or screening for a defined phenotype, followed by propagation and genetic confirmation of candidate mutants. In genetics research, these protocols help link gene changes to observable traits, reveal gene function, and generate experimental models for studying biological pathways. Careful control of growth conditions, appropriate controls, and validation of inheritance improve the reliability of mutant identification and support downstream molecular and phenotypic analyses.

Neutrophils are among the first cells to arrive on the site of inflammatory immune response, and their functions and mechanisms have been studied extensively in vitro. We demonstrate a standard density gradient separation method to isolate human neutrophils from whole blood using commercially available separation media.

IDH1-mutant diffuse low-grade gliomas are rare tumors that are difficult to culture. We introduce a simple, non-enzymatic explant method that preserves IDH1-mutant protein expression and sustains cultures for months. Importantly, this technique works with cryopreserved tumors, offering a valuable resource for glioma research.

Isolating Interaction-Null/Impaired Mutants Using the Yeast Two-Hybrid Assay

The interactions of biomolecules, such as protein-protein interactions, are the molecular basis of biological functions. If interaction-null/impaired mutants that specifically lack the relevant interaction can be isolated, they will greatly help to understand the function(s) of this interaction. This article presents an efficient way to isolate interaction-null/impaired mutants.
