Cfu Quantification Method

The CFU quantification method is a microbiological technique for estimating the number of viable, culturable microorganisms in a sample, making it essential for measuring bacterial or fungal burden. Researchers serially dilute a specimen, spread or pour defined volumes onto growth agar, incubate the plates under suitable conditions, and count the resulting colonies; each colony-forming unit (CFU) represents a viable cell or cell cluster capable of reproduction. In immunology and infection research, CFU counts quantify pathogen loads in cultures, tissues, or experimental models, enabling evaluation of host defense, disease progression, antimicrobial efficacy, and treatment outcomes.

We present a method for quantifying growth phenotypes of individual yeast cells as they grow into colonies on solid media using time-lapse microscopy termed, One-cell Doubling Evaluation of Living Arrays of Yeast (ODELAY). Population heterogeneity of genetically identical cells growing into colonies can be directly observed and quantified.
