Drug-resistant Candida

Drug-resistant Candida refers to Candida yeasts that withstand one or more antifungal drugs, making candidiasis harder to treat and increasing risks for vulnerable patients. Resistance can arise through target-site mutations, increased drug efflux, altered cellular pathways, or biofilm growth, which limits drug penetration and protects fungal communities. Clinicians and researchers use susceptibility testing, genomic analysis, and surveillance to identify resistant strains, select effective therapy, and monitor emerging threats. Studying drug-resistant Candida supports development of new antifungals, improved diagnostic strategies, infection-control practices, and treatment approaches for immunocompromised and hospitalized patients.

Candida albicans Biofilm Chip (CaBChip) for High-throughput Antifungal Drug Screening

We have developed a high-density microarray platform consisting of 3D nano-biofilms of C. albicans called CaBChip. The susceptibility profile of drugs tested on a CaBChip is comparable to the conventional 96-well plate model, suggesting that the fungal chip is ideally suited for true high-throughput screening of antifungal drugs.

Whole Genome Sequencing of Candida glabrata for Detection of Markers of Antifungal Drug Resistance

This study implemented whole genome sequencing for analysis of mutations in genes conferring antifungal drug resistance in Candida glabrata. C. glabrata isolates resistant to echinocandins, azoles and 5-flucytosine, were sequenced to illustrate the methodology. Susceptibility profiles of the isolates correlated with presence or absence of specific mutation patterns in genes.

Drug resistance to targeted therapeutics is widespread and the need to identify mechanisms of resistance--prior to or following clinical onset--is critical for guiding alternative clinical management strategies. Here, we present a protocol to couple derivation of drug-resistant lines in vitro with sequencing to expedite discovery of these mechanisms.
