Modified Mrna Transfection

Modified mRNA transfection is a technique that introduces chemically modified messenger RNA into cells to produce a chosen protein without altering genomic DNA. Delivery systems such as lipid nanoparticles or electroporation transport the mRNA across the cell membrane, where ribosomes translate it in the cytoplasm; modified nucleosides can reduce innate immune sensing and improve protein production. In immunology and infection research, the method enables temporary expression of pathogen antigens, cytokines, or other immune regulators for studying host responses, developing mRNA vaccines, and testing therapeutic strategies. Its transient, programmable activity supports rapid investigation of emerging infectious diseases.

In vitro Transcription and Capping of Gaussia Luciferase mRNA Followed by HeLa Cell Transfection

This method describes high yield in vitro synthesis of both capped and uncapped mRNA from a linearized plasmid containing the Gaussia luciferase (GLuc) gene. The RNA is purified and a fraction of the uncapped RNA is enzymatically capped using the Vaccinia virus capping enzyme. In the final step, the mRNA is transfected into HeLa cells and cell culture supernatants are assayed for luciferase activity.

Gene Transfection toward Spheroid Cells on Micropatterned Culture Plates for Genetically-modified Cell Transplantation

This protocol describes a cell transplantation system using genetically modified, injectable spheroids. Cell spheroids are cultured on micropatterned culture plates and recovered after gene introduction using polyplex nanomicelles. This system facilitates prolonged transgene expression from the transplanted cells in host animals while maintaining the innate function of the cells.
