Monocyte Transfection

Monocyte transfection is the deliberate delivery of nucleic acids into monocytes, innate immune cells that can differentiate into macrophages or dendritic cells, to alter gene expression or produce a desired protein. Because monocytes are sensitive to cellular stress and readily activate inflammatory pathways, successful transfection depends on selecting compatible carriers or electrical methods and carefully controlling exposure conditions to preserve viability and function. In immunology and infection research, transfected monocytes support studies of cytokine regulation, host-pathogen interactions, antigen presentation, and antiviral or antimicrobial responses. This approach can also enable targeted gene knockdown or reporter expression, helping researchers connect specific molecular pathways with immune-cell behavior and evaluate therapeutic strategies.

This study compares two different methods of human monocyte isolation for obtaining in vitro dendritic cells (DCs). Monocytes are selected by adherence or negatively enriched by magnetic separation. We present our optimized high-throughput nucleofection protocol as an efficient way of transfecting primary human monocyte-derived dendritic cells with either plasmid DNA or siRNA without causing cell maturation. We further provide evidence for successful siRNA silencing of targeted gene RIG-I at both the mRNA and protein levels.
