Pathogen Load Measurement

Pathogen load measurement is the quantitative assessment of infectious organisms or their biological material in a defined clinical or experimental sample, providing an indicator of infection burden. Depending on the pathogen and specimen, researchers measure viable organisms by culture, genetic material by quantitative PCR, or pathogen-derived antigens using immunoassays; results are typically reported relative to sample volume, mass, or cell number. In immunology and infection research, these measurements help relate pathogen burden to host immune responses, disease severity, transmission potential, and treatment effectiveness. Repeated measurements can also reveal whether an infection is expanding, controlled, or becoming resistant to therapy.

Single Cell Measurements of Vacuolar Rupture Caused by Intracellular Pathogens

We describe a method for tracking the endomembrane rupture elicited by the intracellular bacteria Shigella flexneri and Mycobacterium tuberculosis upon host cell invasion. Our assay makes use of CCF4, a host cytoplasmic FRET probe in live or fixed cells. This reporter is degraded by an enzyme activity present on the bacterial surface.

Listeria monocytogenes is a model organism for studying immune responses and genetic susceptibility to intracellular bacteria in mice. This method enables one to measure bacterial load and generate single-cell suspensions of the liver and spleen from mice for FACS analysis to determine changes in immune cells due to Listeria infection.
