Pulmonary Macrophages

Pulmonary macrophages are immune cells that reside in the airways and lung tissue, where they protect the respiratory system while helping maintain tissue homeostasis. Alveolar macrophages continuously sample inhaled material, engulf microbes and particles through phagocytosis, and regulate inflammation by releasing cytokines and clearing damaged cells and excess surfactant. Their responses depend on signals from airway epithelium, lung fluids, and invading pathogens, allowing them to balance antimicrobial defense with prevention of excessive tissue injury. Studying pulmonary macrophages is essential for understanding respiratory infections, asthma, chronic inflammatory diseases, and host responses to inhaled therapies and environmental exposures.

Investigation of Macrophage Polarization Using Bone Marrow Derived Macrophages

The article describes a readily easy adaptive in vitro model to investigate macrophage polarization. In the presence of GM-CSF/M-CSF, hematopoietic stem/progenitor cells from the bone marrow are directed into monocytic differentiation, followed by M1 or M2 stimulation. The activation status can be tracked by changes in cell surface antigens, gene expression and cell signaling pathways.

The protocol presented here describes methods for depleting macrophages in vivo, deriving polarized macrophages from bone marrow aspirates, and adoptively transferring macrophages into mice.
