Recombinant Virus Production

Recombinant virus production is the laboratory process of engineering viral genomes and generating virus particles that carry selected genetic sequences for research, therapeutic, or vaccine applications. It typically involves inserting a target gene into a viral vector, introducing the engineered genome into permissive producer cells, and allowing cellular machinery to assemble and release recombinant particles under controlled conditions. Researchers then characterize particle yield, genetic payload, functional activity, and biosafety before use. In immunology and infection research, these viruses help deliver antigens, model host–pathogen interactions, measure immune responses, and evaluate vaccines or gene-delivery strategies.

Here we describe a protocol based only on cell infection, which improves the efficiency of recombinant parvovirus production by more than 100 fold in comparison to other protocols in use. This protocol relies on the use of a novel adenovirus 5-based helper containing the parvovirus VP transcription unit (Ad-VP).

Generating Recombinant Live-Attenuated Influenza Viruses for Vaccines
