Super Speed Centrifugation

Super speed centrifugation is a laboratory separation technique that uses high rotational speeds to fractionate particles, cells, or biomolecules according to their size, density, and shape. During centrifugation, centrifugal force drives denser components through a liquid medium, forming a pellet while less dense material remains in the supernatant; rotor speed, time, and sample composition influence the separation. In immunology and infection research, the method can concentrate microbial particles, enrich viral or cellular fractions, and prepare samples for downstream analysis of pathogens, antigens, antibodies, or immune responses. These applications support pathogen characterization, sample purification, and investigation of host–microbe interactions.

Recently we mapped the three-dimensional (3D) spatial locations of transport routes for various proteins translocating inside primary cilia in live cells. Here this paper details the experimental setup, the process of biological samples and the data analyses for the 3D super-resolution fluorescence imaging approach newly applied in live primary cilia.

Cell Fractionation of U937 Cells in the Absence of High-speed Centrifugation

Here, we present a protocol to isolate the plasma membrane, cytoplasm and mitochondria of U937 cells without the use of high-speed centrifugation. This technique can be used to purify subcellular fractions for subsequent examination of protein localization via immunoblotting.
