Cardiac Macrophage Isolation

Cardiac macrophage isolation is a laboratory technique used to separate macrophages from heart tissue for studying their roles in cardiac health and disease. The process typically involves enzymatic dissociation of the tissue into a cell suspension, followed by filtration, centrifugation, and selection based on macrophage surface markers or physical properties. Isolated cells can then be characterized by flow cytometry, microscopy, or molecular assays to assess inflammatory signaling, tissue repair, and interactions with cardiomyocytes and other immune cells. This approach supports research into myocardial injury, cardiac remodeling, fibrosis, and potential immune-targeted therapies.

This method for isolating functional immune cells from the heart provides an alternative to the conventional methods of collagenase digestion, which causes unwanted immune cell activation, resulting in a decreased responsiveness of these cells. Our method of isolation yields functional cardiac immune cells by avoiding problems associated with enzymatic digestion.

Here, we detail a methodology for the rapid isolation of mouse intestinal dendritic cells (DCs) and macrophages. Phenotypic characterization of intestinal DCs and macrophages is performed using multi-color flow cytometric analysis while magnetic bead enrichment followed by cell sorting is used to yield highly pure populations for functional studies.

(2017)This video demonstrates the magnetic isolation of phagosomes containing biotin–streptavidin bead–coated bacteria from infected macrophages. The protocol includes buffer treatments, cell disruption, magnetic separation, and washing to yield purified phagosomes for analysis.
