Pbmc Pellet Preparation

PBMC pellet preparation is a laboratory method for collecting and concentrating peripheral blood mononuclear cells (PBMCs) into a compact cell fraction for downstream medical and biomedical analyses. The process uses density-based separation or another validated isolation workflow, followed by removal of supernatant, washing to reduce residual plasma and reagents, and controlled centrifugation to form a pellet that can be resuspended in an appropriate medium. Consistent pellet preparation supports cell counting, viability assessment, immunophenotyping, molecular assays, and cell culture, while careful handling helps preserve cellular integrity and reproducibility in studies of immunity, infection, inflammation, and patient-derived biomarkers.

This video demonstrates an assay to generate a humanized xenograft mouse model. An immunocompromised mouse is injected with cyclophosphamide to cause immune cell depletion. Patient-derived tumor cells mixed with human peripheral blood mononuclear cells (PBMCs) in a hydrogel are subcutaneously injected into the mouse, forming a tumor with infiltrating T cells.

A rapid bacterial pellet preparation from a positive blood culture can be used as a sample for applications such as identification by MALDI-TOF, Gram staining, antibiotic susceptibility testing and PCR-based test. The results can be rapidly communicated to clinicians to improve the outcome of patients suffering from bloodstream infections.

Method to Produce Durable Pellets at Lower Energy Consumption Using High Moisture Corn Stover and a Corn Starch Binder in a Flat Die Pellet Mill

In this study, a protocol was developed to produce good quality pellets using a flat die pellet mill at reduced specific energy consumption testing high-moisture corn stover and a starch based binder. The results indicated that adding a corn starch binder improved the pellet durability, reduced percent fines and decreased specific energy consumption.
