Serial Transverse Sectioning

Serial transverse sectioning is a histological method that produces a sequence of thin, parallel slices across the short axis of a tissue or organ, enabling its three-dimensional structure to be examined from two-dimensional sections. After fixation and embedding, the specimen is cut at successive depths, with each section collected, mounted, and often stained for microscopic analysis. In medicine, this approach reveals how anatomical features, lesions, vessels, and cellular patterns change through a specimen’s depth. Aligning consecutive sections can also support three-dimensional reconstruction, improving the study of pathology, developmental anatomy, and spatial relationships within complex tissues.

This protocol targets specific cells in tissue for imaging at nanoscale resolution using a scanning electron microscope (SEM). Minimally invasive transverse aortic constriction (MTAC) conserves the essentials of regular transverse aortic constriction (TAC) while eliminating the use of a ventilator with tracheal intubation. It proves to be a highly desirable method for high-throughput studies on left ventricular overload, particularly in translational studies.
