Three-dimensional Organization

Three-dimensional organization is the spatial arrangement of cells, tissues, molecules, and structures within a volume, a feature that shapes how biological systems function and respond to disease. It emerges through coordinated cell-cell interactions, extracellular matrix signaling, tissue geometry, and chemical or mechanical gradients that establish local architecture and communication. In medicine, understanding this organization improves interpretation of tissue pathology and supports the development of organoids, three-dimensional cell cultures, imaging methods, and tissue-engineering strategies. Preserving realistic spatial relationships can also strengthen disease modeling, drug evaluation, and regenerative medicine by producing results that more closely reflect human physiology.

Culture of normal cells in their three-dimensional context represents an alternative method to study early events required for cellular transformation and tumorigenesis. This method is used to grow normal ovarian and oviductal cells to study early events in ovarian cancer formation.

A Whole-Mount Organ Staining Technique for Three-Dimensional Imaging of the Kidney

2025

This video demonstrates an assay for the whole-mount staining of murine kidneys using a tissue-clearing method called CUBIC (clear, unobstructed brain/body imaging cocktails and computational analysis) and immunofluorescent staining. The kidney is subjected to tissue clearing, wherein the kidney is delipidated, decolorized, and treated with a refractive index matching solution to make the tissue transparent for optimum three-dimensional imaging. The tissue is stained with fluorescently labeled...
