Calcium Binding Dye

Calcium binding dyes are chemical indicators that detect changes in free calcium ion concentration by producing a measurable change in fluorescence or color. Their indicator molecules bind Ca2+ selectively, altering the dye’s optical properties, so researchers can monitor calcium dynamics in living cells with fluorescence microscopy. In neuroscience, these dyes help reveal neuronal activity because action potentials and synaptic signaling can trigger transient increases in intracellular calcium. Calcium imaging with such indicators supports studies of neuronal communication, circuit function, and cellular responses, while careful control of dye loading, concentration, and imaging conditions helps preserve cell health and improve measurement accuracy.

Assessing T-cell Receptor-Induced Calcium Influx Using a Calcium Indicator Dye

This video analyzes T-cell receptor-induced calcium influx in T-cells. Anti-CD3 antibodies activate the T-cell signaling pathways, leading to an increase in intracellular calcium ions that bind with Indo-1 dye. In flow cytometry, a spectrum shift from the calcium-free state to the calcium-bound state correlates with T-cell receptor-induced calcium influx.

Loading a Calcium Dye into Frog Nerve Endings Through the Nerve Stump: Calcium Transient Registration in the Frog Neuromuscular Junction

Here, we describe a method for loading a calcium-sensitive dye through the frog nerve stump into the nerve endings. We also present a protocol for the recording and analysis of fast calcium transients in the peripheral nerve endings.

Calcium-Dependent Hydrophobic Interaction Chromatography: A Technique to Purify Calcium-Binding Proteins Based on Hydrophobic Interactions

In this video, we demonstrate the purification of calcium-binding protein from a dialyzed cell lysate through calcium-dependent hydrophobic interaction chromatography. The calcium-binding proteins expose a hydrophobic region upon binding with calcium, facilitating interaction with a hydrophobic group on resin. Later these proteins are eluted using calcium chelator EDTA that reverses the interaction.

Direct Imaging of ER Calcium with Targeted-Esterase Induced Dye Loading (TED)

Targeted-esterase induced dye loading (TED) supports the analysis of intracellular calcium store dynamics by fluorescence imaging. The method bases on targeting of a recombinant Carboxylesterase to the endoplasmic reticulum (ER), where it improves the local unmasking of synthetic low-affinity Ca2+ indicator dyes in the ER lumen.
