Focal Cortical Lesion

A focal cortical lesion is a localized area of structural damage or abnormality within the cerebral cortex, a region that supports perception, movement, language, memory, and other higher functions. By disrupting cortical neurons and their connections, the lesion can alter local signaling and sometimes increase network excitability, producing symptoms that depend on its location, size, and underlying cause. In neuroscience, focal cortical lesions provide important models for linking brain regions to behavior and cognition. Clinical evaluation commonly combines neurological examination with magnetic resonance imaging, electroencephalography, and lesion-symptom mapping to guide diagnosis, monitor progression, and inform treatment, including management of seizure-related disorders.

Photothrombosis is a quick, minimally-invasive technique for inducing small and well-delimited infarction in areas of interest in highly reproducible manner. It is particularly suitable for studying cellular and molecular responses underlying brain plasticity in transgenic mice.

A high resolution ex vivo 7T MR imaging protocol is presented, to perform MR-guided histopathological validation of microvascular pathology in post-mortem human brain tissue. Furthermore, guidelines are provided for the assessment of cortical microinfarcts on in vivo 7T as well as 3T MR images.

This protocol describes a custom overpressure air system designed to induce closed-system central nervous system (CNS) injuries in mice, including ocular, brain, and spinal cord traumas. Experimental Demyelination and Remyelination of Murine Spinal Cord by Focal Injection of Lysolecithin

Demyelinating diseases can be modeled in animals by focal application of lysolecithin into the CNS. A single injection of lysolecithin into mouse spinal cord produces a lesion that spontaneously repairs over time. The goal is to study factors involved in de- and remyelination, and to test agents for enhancing repair.
