Human Induced Pluripotent Stem Cells

Human induced pluripotent stem cells (hiPSCs) are mature human cells reprogrammed to an embryonic-like, pluripotent state, making them a versatile model for studying human biology and disease. Typically, defined transcription factors are introduced into somatic cells to reset gene-expression and epigenetic programs, after which the cells can self-renew and differentiate into neural progenitors and diverse neuronal or glial cell types. In neuroscience, patient-derived hiPSCs enable investigation of disease-associated cellular phenotypes in a human genetic context, including disorders that are difficult to model in animals. They also support drug screening, toxicity testing, and research on neural development and regenerative strategies.

Despite recent advancements in genetic modification, transfection of human embryonic stem cells (HESCs) remains a capricious process. To our knowledge, systematic and efficient methods to transfect human induced pluripotent stem cells (iPSCs) have not been reported. Here, we describe robust protocols to efficiently transfect and nucleofect human iPSCs.

A method to generate human induced pluripotent stem cells (iPSCs) via retrovirus-mediated ectopic expression of OCT4, SOX2, KLF4 and MYC is described. A practical way to identify human iPSC colonies based on GFP expression is also discussed.

This video describes an in vitro method for generating macrophages from human induced pluripotent stem cells (iPSCs). Macrophage generation from iPSCs occurs in three stages: (i) aggregation of iPSCs in suspension to form three-dimensional embryoid bodies (EBs) that differentiate into mesodermal cells, (ii) formation of macrophage precursor cells from the differentiated EBs, and (iii) differentiation of the macrophage precursor cells into mature, functional macrophages.

Robust derivation of human induced pluripotent stem (hiPS) cells was achieved by using non-integrating Sendai virus (SeV) vector mediated reprogramming of dermal fibroblasts. hiPS cell maintenance and clonal expansion was performed using xeno-free and chemically defined culture conditions with recombinant human laminin 521 (LN-521) matrix and Essential E8 (E8) Medium.
