Neuronal Cell Aggregates

Neuronal cell aggregates are three-dimensional clusters of neurons that recreate aspects of the cellular organization and interactions found in nervous tissue. They form when neurons are maintained under conditions that promote cell-cell adhesion and coordinated signaling, allowing cells to assemble into compact structures rather than remain as isolated cultures. In neuroscience, these aggregates provide a model for studying neuronal development, network formation, synaptic communication, and responses to injury or disease. Their three-dimensional organization can complement conventional monolayer cultures and support investigations of neurotoxicity, disease mechanisms, and potential therapeutic effects in a controlled laboratory setting.

Immunostaining of Neurons Treated with Alpha-Synuclein Aggregates

This video demonstrates the immunostaining of neurons treated with alpha-synuclein (αS) aggregates. The neurons are first treated with αS aggregates. They are then fixed, permeabilized, and treated with a blocking solution. Primary antibodies are added, followed by a fluorescently labeled secondary antibodies and a nuclear stain. The sample is then mounted and observed under a microscope.

4D Imaging of Protein Aggregation in Live Cells

Cellular viability depends on timely and efficient management of protein misfolding. Here we describe a method for visualizing the different potential fates of a misfolded protein: refolding, degradation, or sequestration in inclusions. We demonstrate the use of a folding sensor, Ubc9ts, for monitoring proteostasis and aggregation quality control in live cells using 4D microscopy.
